No, she probably wouldn't get offended, most transgendered people I know don't. They are used to people stumbling around. Just recently a friend of mine who is postop and has been for some time was called he by someone who was signing her in for something. She barely reacted and I quickly and quietly corrected the person under my breath. She smiled thanks at me, and it was forgotten.

Most people don't really get offended when you say something through ignorance or confusion, as opposed to intending to be mean. As you said your joke was as much about your fumbling as anything else. And it was a reasonable fumbling.
